Fluorescent Microscope is used for capillary examination. This technique is a new, highly sensitive rapid method of diagnosing malaria. It is also useful in the diagnosis of filariasis and leptospirosis. The principle of this method is the malaria parasite picks up fluorescent stain into their nucleus and cytoplasm, so that its morphologic characteristics can be examined by fluorescent microscopy.
The nucleus appears green and the cytoplasm reddish orange, The Capillary is coated with Acridine orange on its inside. Parents Blood is loaded into this capillary and centrifuged at 12000 rpm for 5 minutes. The Blood components settle at different levels in the capillary. These different layers are displayed in the picture.
Examination of the area just below granulocyte-RBC junction helps to detect the malarial parasites, as these are concentrated in this are upto 1000 times. Examination of Lymphocyte / monocyte layer using transmitted light helps to detect the schizonts, and gametocytes which gets concentrated in this layer very easily.

Method of Examination:
The granulocyte layer is then seen under 62x oil objective. By changing the light source to fluorescent light the parasitic forms can be seen as greenish orange stained structures. Using only transmitted light the parasitic forms are seen as block pigmented structures as shown in the preceding photomicrographs.

This a new method for identifying the malaria parasite in the peripheral blood. It involves staining of the centrifuged and compressed red cell layer with acridine orange and its examination under UV light source. It is fast, easy and claimed to be more sensitive than the traditional thick smear examination.
Method: The tube is a high-precision glass hematocrit tube, pre-coated internally with acridine orange stain and potassium oxalate. It is filled with 55-65 micro liters of blood from a finger, ear or heel puncture. A clear plastic closure is then attached. A precisely made cylindrical float, designed to be suspended in the packed red blood cells, is inserted. The tube is centrifuged at 12,000 rpm for 5 minutes. The components of the buffy coat separate according to their densities, forming discrete bands. Because the float occupies 90% of the internal lumen of the tube, the leukocyte and the thrombocyte cell band widths and the top-most area of red cells are enlarged to 10 times normal. The tube is placed on the tube holder and examined using a standard white light microscope equipped with the UV microscope adapter, an epi-illuminated microscope objective. Fluorescing parasites are then observed at the red blood cell/white blood cell interface. The key feature of the method is centrifugation and thereby concentration of the red blood cells in a predictable area of the tube, making detection easy and fast.
Red cells containing Plasmodia are less dense than normal ones and concentrate just below the leukocytes, at the top of the erythrocyte column.
The float forces all the surrounding red cells into the 40 micron space between its outside circumference and the inside of the tube. Since the parasites contain DNA which takes up the acridine orange stain, they appear as bright specks of light among the non-fluorescing red cells.
Virtually all of the parasites found in the 60 micro liter of blood can be visualized by rotating the tube under the microscope. Inverted Tissue Culture Microscope is designed to observe tissue cultures, protozoans, planktons and any type of microbes in liquid media in cavity slides, cavity blocks, watch glasses or petri dishes etc. Involving wide range of applications.
Mounted on a pole and can be positioned in vertical height range upto 140 mm. Complete with koehler’s illumination system with focused condenser lens with iris-diaphragm filter-carrier etc.

Metallurgical Microscope has bright field vertical illuminator with high intensity low voltage lamp 6V- 15W with adjustable field and aperture, iris-diaphragm with slots for dropping coloured filters.
It provides excellent bright field in incident illumination of opaque and semi-opaque specimen. The same can be used as normal Pathological microscope. Provided with triple revolving nose piece, built-in co-axial mechanical stage. Supplied in Plywood box.

POLARIZING MICROSCOPES : JS-41

Polarising Microscopes are standard polarising microscopes equipped with strain-free plan achromatic objectives for transmitted light observation. Built-in halogen illumination system and high quality coated optics make the equipment versatile and high quality instrument.
Polarising intermediate has attachment magnification of 1.2X and has built-in Bertrand lens unit on a rotating turret for colonoscopy, a swing in / swing out analyser and DIN standard compensatory slot accommodating 1st order red sensitive tint plate and ¼ wave length radiation plate.
The microscope stand has a quadruple ball bearing nose-piece, co-axial coarse and fine motions moving on ball bearing guide. Sub-stage polariser on strain-free Abbe condenser Numerical Aperture of 1.25.
